Anti-infectious activity of human mannose-binding lectin (MBL)

Abstract: Mannose-binding lectin (MBL) is an important constituent of the innate immune system. It belongs to the collectin family of proteins which are characterized by the presence of both a collagenous region and a carbohydrate recognition domain. In humans the proteins of this family are encoded by a cluster of genes located on the long arm of the chromosome 10. Low levels of MBL are caused by single nucleotide polymorphisms within exon l or in the promoter region of the MBL-2 gene. Such mutations have been shown to be associated with an increased susceptibility to many infections. Absence or extremely Iow concentration of serum MBL seems to be a risk factor for occurrence of some disorders, in particular autoimmune diseases. MBL can bind through multiple sites to a range of sugars including mannose, N-acetylglucosamine and interact with a wide spectrum of bacteria, viruses, yeasts, fungi, and protozoa decorated with such sugars. MBL bound to microbial surfaces is able to activate complement system via the lectin pathway. This protein may also interact directly with cell surface receptors and thereby promote opsonophagocytosis by a complement-independent pathway. There have been a number reports that MBL can modulate the inflammatory response, but it is likely that the naturę of the response is different for various organisms or group of organisms.
