Functions and sorting of Gram-negative bacteria lipoproteins
Abstract: Lipid modified proteins, which belong to a group of membrane proteins, are present in cells of various organisms, from bacteria to eukaryotes. Lipid modification of these proteins occurs post-translationally on the periplasmic surface of the inner membrane. Lipoproteins of Gram-negative bacteria are anchored into inner or outer membrane via fatty acids attached to N-terminal cysteine. These proteins perform many various functions in bacterial cells. They may interact with TLR receptors and stimulate several signal transduction pathways leading to cytokine genes expression. Lipoproteins are transported by CM by the Sec protein machinery. The membrane localization of the
E. coli lipoproteins is dependent of the lipoprotein sorting signal, the amino acid residue next to the lipid modified cysteine. Generally an Asp at this position resulted in retention of the lipoprotein in inner membrane, whereas others residues direct lipoproteins to the outer membrane. In
E. coli LolA-LolB system, consisting of five proteins (LolABCDE), is the general mechanism by which outer membrane-directed lipoproteins are efficiently released from cytoplasmic membrane and transported into outer membrane.
